The Swedish international, who has been in fine form with three goals in his last four league games, appeared to suffer a hip injury following a collision with Tyrick Mitchell.
Isak initially attempted to continue after treatment but struggled with movement and eventually signaled that he could not proceed.
Manager Eddie Howe replaced him with Harvey Barnes, adjusting the lineup by moving Anthony Gordon to a central role and deploying Barnes on the left wing.
Injury Woes for Newcastle
The match highlighted Newcastle’s ongoing injury struggles, with Joe Willock and Joelinton also requiring medical attention during the first half.
Despite fitness concerns, Joelinton and Bruno Guimarães were included in the starting lineup, but their readiness seemed questionable.
While Newcastle managed their injuries, it was Crystal Palace who dominated early chances. Nick Pope denied Ismaila Sarr with a close-range save, and Daniel Muñoz squandered a prime opportunity, firing wide from six yards out following a sharp counterattack.
